Molecular cloning, characterization and constitutive expression analysis of TLR4 gene in Pangasianodon hypophthalmus

Toll like receptor (TLR) signalling pathway plays a central role in inflammatory responses through the recognition of PAMPs and DAMPs. Toll-like 4 (TLR4) is well known for LPS gram-negative bacteria leading to production cytokines. Modulation TLR gene different tissues indicates functional importance innate immunity. The aim this study was clone characterize TLR4 evaluate its constitutive tissue distribution Pangasianodon hypophthalmus, commercially important fish species. carried out at ICAR-CIFE, Mumbai during 2017. Partial nucleotide amino acid sequences were identified P. hypophthalmus termed as phTLR4. Bio-informatics analysis partial sequence revealed four putative LRR domains characteristic TLRs. Nucleotide showed varying degree identity with from other species BLASTn BLASTp analysis. Phylogenetic placed phTLR4 close Ictalurus punctatus. found be constitutively expressed all studied. significantly highest expression recorded intestine spleen. lowest muscle, heart skin. House-keeping ß-actin used an internal control. outcome research will helpful studying mRNA levels under conditions hypophthalmus.
